Treating DNA with bisulfite chemically modifies unmethylated cytosines into uracil, and methylated cytosines remain unchanged. Once converted, the methylation profile of the DNA can be determined using the desired downstream application (Methylation-specific PCR, Sanger sequencing, Pyrosequencing®, array-based methods, reduced representation bisulfite sequencing (RRBS), and whole genome bisulfite sequencing.
Bisulfite conversion is the gold standard technology for DNA methylation analysis, providing single nucleotide resolution.
